Abstract
Gliclazide is a new sulfonylurea hypoglycemic agent which lias been studied for its clinical and -hortterm tolerance to 8 newly diagnosed patients (Group I) and 20 unsuccessfully treated patients ( 0 roup II) with other sulfonylureas.
The results obtained were as follows:
1. The mean duration of drug use in group 1(4.2¡¾1.8 weeks)was shorter than that of group II (9.¢¥ ¡¾7.2 weeks).
2. The numbers (%) of patients whose blood sugars were controlled well were 8 (100%) in group I and 10 (50%) in gorup II, and overall response rate was 64.2%.
3. The mean dosages of gliclazide used in group I and group II were 120¡¾86.4 mg/day and 176 63.2 mg/day, respectively.
4. The mean fasting blood sugars before gliclazide in group I (258.5¡¾45.9 mg/dl) and group 11(199.7 42.6 mg/dl) lowered by 57.3% and 31.2% respectively after gliclazide. The mean postprandial 2hr 11lood sugars before gliclazide in group 1 (378.2 ¡¾ 17.2 mg/dl) axid group 11 (242.2 ¡¾332;9 mg/dl) lower: d by 59.4% and 27.4% respectively after gliclazide.
5. There were positive correlation between the pre-treatment obesity of patients and weight loss .fter gliclazide treatment.
6. During treatment one patient experienced transient epigastric discomfort and another one expe ¢¥enced mild headache. Two patients also felt mild hypoglycemic symptom for a moment which disapp=. ed after dose adjustment.
These results suggested that gliclazide is a very effective and safe oral hypoglycemic agent in t eatment of non-insulin dependent diabetic patients.
